چکیده

Initial attempts to grow N2-fixing Derxia gummosa autotrophically on H2, C 0 2 , O2 and N2 in a closed system yielded variable results. Poor growth was found to be due to rapid O2 depletion and the requirement for an agar surface. In a closed system, C2H2 reduction assays could not be carried out due to complete consumption of H2. Hence a flow-through culturing technique was developed to supply gases at a constant partial pressure and to perform C2H2 reduction assays in a continuous flow system. Hydrogenase of autotrophic D. gummosa was not inhibited by C2H2, even at 0.5 atm, and the K,,, of hydrogenase for H2 was approximately 0.15 atm. The effects of O2 and H2 on C2Hz reduction were examined, using the flow-through assay system. The rate of C2H2 reduction decreased below 0.074 atm H2, suggesting that ATP and reductant supply were limiting the nitrogenase activity.

Comparative Cytochrome Oxidase and Superoxide Dismutase Analyses on Strains of Azotobacter vinelandii and Other Related Free-Living Nitrogen-Fixing Bacteria.

Quantitative N,N,N',N'-tetramethyl-p-phenylenediamine (TMPD) oxidase and superoxide dismutase (SOD) analyses were performed on representative organisms of the family Azotobacteraceae. Azotobacter vinelandii, Azotobacter chroococcum, Azotobacter paspali, and Derxia gummosa exhibited high quantitative TMPD oxidase activities, and their extracts possessed very active and electrophoretically homoge...

Chemoautotrophic growth of hydrogen-uptake-positive strains of Rhizobium japonicum.

Recently reported research from this laboratory has demonstrated the autotrophic growth of certain hydrogen-uptake-positive strains of Rhizobium japonicum and defined minimal conditions for such growth. Ribulose 1,5-bisphosphate carboxylase has been detected in autotrophically growing cells, but at low specific activity. Moreover, growth rates were low, and growth ceased at low cell densities. ...

Nitrogen Fixation Aligns with nifH Abundance and Expression in Two Coral Trophic Functional Groups

Microbial nitrogen fixation (diazotrophy) is a functional trait widely associated with tropical reef-building (scleractinian) corals. While the integral role of nitrogen fixation in coral nutrient dynamics is recognized, its ecological significance across different coral functional groups remains yet to be evaluated. Here we set out to compare molecular and physiological patterns of diazotrophy...

The Rnf Complex of Clostridium ljungdahlii Is a Proton-Translocating Ferredoxin:NAD+ Oxidoreductase Essential for Autotrophic Growth

UNLABELLED It has been predicted that the Rnf complex of Clostridium ljungdahlii is a proton-translocating ferredoxin:NAD(+) oxidoreductase which contributes to ATP synthesis by an H(+)-translocating ATPase under both autotrophic and heterotrophic growth conditions. The recent development of methods for genetic manipulation of C. ljungdahlii made it possible to evaluate the possible role of the...
